Sphingosines affect the adherence of Streptococcus gordonii and Streptococcus sanguinis on hydroxyapatite

چکیده
Sphingosines affect the adherence of Streptococcus gordonii and Streptococcus sanguinis on hydroxyapatite. Abstract Sphingosines exhibit bactericidal activity on planktonic cells. However, their anti-adherence activity when adsorbed on hydroxyapatite (HA) surfaces is unknown. Therefore this study evaluated the anti-adherence activity of a variety of sphingosines and their derivatives against Streptococcus gordonii and Streptococcus sanguinis. HA discs were treated with sphingosines, and subsequently incubated with S. gordonii or S. sanguinis. The number of surface-adherent bacteria was evaluated by culturing and microscopy. The anti-adherence activity of sphinganine was evaluated on 24 h old biofilms in the presence and absence of sucrose. Among the tested sphingosines, sphinganine inhibited adherence of S. gordonii and S. sanguinis by 80-and 100-fold, respectively. Growth of S. gordonii was inhibited 75-fold in the presence and absence of sucrose, while the growth of S. sanguinis was inhibited only in the absence of sucrose. These results suggest that sphinganine display potential as coating material to prevent biofilm formation on HA.

Streptococcus gordonii Hsa environmentally constrains competitive binding by Streptococcus sanguinis to saliva-coated hydroxyapatite.
Competition between pioneer colonizing bacteria may determine polymicrobial succession during dental plaque development, but the ecological constraints are poorly understood. For example, more Streptococcus sanguinis than Streptococcus gordonii organisms are consistently isolated from the same intraoral sites, yet S. gordonii fails to be excluded and survives as a species over time. To explain ...
متن کاملStreptococcal antagonism in oral biofilms: Streptococcus sanguinis and Streptococcus gordonii interference with Streptococcus mutans.
Biofilms are polymicrobial, with diverse bacterial species competing for limited space and nutrients. Under healthy conditions, the different species in biofilms maintain an ecological balance. This balance can be disturbed by environmental factors and interspecies interactions. These perturbations can enable dominant growth of certain species, leading to disease. To model clinically relevant i...
متن کاملCharacterization of hydrogen peroxide-induced DNA release by Streptococcus sanguinis and Streptococcus gordonii.
Extracellular DNA (eDNA) is produced by several bacterial species and appears to contribute to biofilm development and cell-cell adhesion. We present data showing that the oral commensals Streptococcus sanguinis and Streptococcus gordonii release DNA in a process induced by pyruvate oxidase-dependent production of hydrogen peroxide (H(2)O(2)). Surprisingly, S. sanguinis and S. gordonii cell int...

Objectives: This study aimed to assess the antimicrobial effects of aloe vera hydroalcoholic extract and 0.2% chlorhexidine (CHX) mouthwash on Streptococcus sanguinis (S. sanguinis), Streptococcus salivarius (S. salivarius) and Streptococcus mutans (S. mutans) in vitro. Materials and Methods: Four concentrations (25%, 50%, 75% and 100%) of hydroalcoholic extract of aloe vera were prepared. S. ...
متن کاملRole of Streptococcus sanguinis sortase A in bacterial colonization.
Streptococcus sanguinis, a normal inhabitant of the human oral cavity, has low cariogenicity, though colonization on tooth surfaces by this bacterium initiates aggregation by other oral bacteria and maturation of dental plaque. Additionally, S. sanguinis is frequently isolated from infective endocarditis patients. We investigated the functions of sortase A (SrtA), which cleaves LPXTG-containing...
